Understanding the distribution of elements within plant tissues is important across a range of fields in plant science. In this review, we examine synchrotron-based x-ray fluorescence microscopy (XFM) as an elemental imaging technique in plant sciences, considering both its historical and current uses as well as discussing emerging approaches. Synchrotron x-ray imaging was performed at the Advanced Photon Source (APS) of the Argonne National Laboratory, yielding images of high spatial (~2 μm) and temporal (~20,000 Hz) resolution. Clear images were obtained using a camera with a high frame rate. The x-ray source is described in the Supplementary Materials. Synchrotron soft x-ray techniques are already opening up new ways to address the complex problems arising from earth resource utilisation, and this contribution is expected to increase in areas such as environmentally sustainable ore extraction, mineral processing, coal combustion and soil use. Synchrotron radiation is the brightest artificial source of X-rays. The planar acceleration geometry appears to make the radiation linearly polarized when observed in the orbital plane , and circularly polarized when observed at a small angle to that plane.

The X-ray diffraction (XRD) end station measures constructive interference of the x-ray wave with repeating atomic and interfacial structure in materials. At the Beamline for Material Measurement, XRD features a tunable, monochromatic, high intensity beam with a beam spot on the order of 100 micrometers.
